Gut microbiota induced epigenetic modifications in the non‐alcoholic fatty liver disease pathogenesis

چکیده
Non-alcoholic fatty liver disease (NAFLD) represents a growing global health concern that can lead to and cancer. It is characterized by an excessive accumulation of fat in the liver, unrelated alcohol consumption. Studies indicate gut microbiota-host crosstalk may play causal role NAFLD pathogenesis, with epigenetic modification serving as key mechanism for regulating this interaction. In review, we explore how interplay between microbiota host epigenome impacts development NAFLD. Specifically, discuss microbiota-derived factors, such lipopolysaccharides (LPS) short-chain acids (SCFAs), modulate DNA methylation histone acetylation genes associated NAFLD, subsequently affecting lipid metabolism immune homeostasis. Although current literature suggests link development, our understanding molecular mechanisms signaling pathways underlying remains limited. Therefore, more comprehensive epigenomic multi-omic studies, including broader clinical animal experiments, are needed further linking NAFLD-associated genes. These studies anticipated improve microbial markers based on strategies provide novel insights into pathogenesis ultimately addressing significant unmet need.

Gut microbiota and nonalcoholic fatty liver disease
Recent evidence has linked obesity and the metabolic syndrome with gut dysbiota. The precise mechanisms underlying that association are not entirely understood; however, microbiota can enhance the extraction of energy from diet and regulate whole-body metabolism towards increased fatty acids uptake from adipose tissue and shift lipids metabolism from oxidation to de novo production. Obesity and...

The 5-Hydroxymethylcytosine (5-hmC) is an epigenetic modification whose role in the pathogenesis of metabolic-related complex diseases remains unexplored; 5-hmC appears to be prevalent in the mitochondrial genome. The Ten-Eleven-Translocation (TET) family of proteins is responsible for catalyzing the conversion of 5-methylcytosine to 5-hmC. We hypothesized that epigenetic editing by 5-hmC might...
متن کاملImplication of Gut Microbiota in Nonalcoholic Fatty Liver Disease
It is now well established that gut flora and chronic liver diseases are closely interrelated. This association is most evident at late stages of the disease: cirrhosis and impaired liver function are associated with intestinal bacterial overgrowth, small bowel dysmotility, increased gut permeability, and decreased immunological defenses, all of which promote bacterial translocation from the gu...

متن کاملThe Pathogenesis of Nonalcoholic Fatty Liver Disease: Interplay between Diet, Gut Microbiota, and Genetic Background.
Nonalcoholic fatty liver disease (NAFLD) is the most common chronic liver disease in the world, and it comprises a spectrum of hepatic abnormalities from simple hepatic steatosis to steatohepatitis, fibrosis, cirrhosis, and liver cancer. While the pathogenesis of NAFLD remains incompletely understood, a multihit model has been proposed that accommodates causal factors from a variety of sources,...
